Men and women can't be friends because sex always gets in the way. College graduates Harry (Billy Crystal) and Sally (Meg Ryan), who have just met, argue about the truth of this theory during an 18-hour car journey to New York. When they arrive at their destination, they go their separate ways. Ten years and two failed relationships later, they meet again by chance in a bookshop. The two decide to prove them wrong and, against all expectations, become best friends - but they find it increasingly difficult to maintain their purely platonic relationship.
